RESUMO

Norovirus (NoV) stands as a significant causative agent of nonbacterial acute gastroenteritis on a global scale, presenting a substantial threat to public health. Hence, the development of simple and rapid analytical techniques for NoV detection holds great importance in preventing and controlling the outbreak of the epidemic. In this work, a self-powered photoelectrochemical (PEC) immunosensor of NoV capsid protein (VP1) was proposed by the π-electron-rich carbon nitride homojunction (ER-CNH) as the photoanode. C4N2 ring derived from π-rich locust bean gum was introduced into the tri-s-triazine structure, creating a large π-delocalized conjugated carbon nitride homojunction. This strategy enhances the C/N atomic ratio, which widens light utilization, narrows the bandgap, and optimizes the electronic band structure of carbon nitride. By introduction of a π-rich conjugated structure, p-type domains were induced within n-type domains to build the internal electric field at the interface, thus forming a p-n homojunction to boost carrier separation and transfer. The ER-CNH photoanode exhibited excellent photoelectric performance and water oxidation capacity. Since VP1 inhibits the water oxidation of the ER-CNH photoanode, the open-circuit potential of the as-prepared PEC immunosensor system was reduced for detecting NoV VP1. The self-powered PEC immunosensor achieved a remarkably low detection limit (∼5 fg mL-1) and displayed high stability and applicability for actual stool samples. This research serves as a foundation concept for constructing immunosensors to detect other viruses and promotes the application of self-powered systems for life safety.

RESUMO

Syndecan-1 (SDC1) modified lipid bilayer (LB)-coated mesoporous silica nanoparticles (MSN) to co-deliver gemcitabine (GEM) and honokiol (HNK) were prepared for the targeting treatment of pancreatic cancer. The encapsulation efficiencies of GEM and HNK in SDC1-LB-MSN-GEM/HNK were determined to be 60.3 ± 3.2% and 73.0 ± 1.1%. The targeting efficiency of SDC1-LB-MSN-GEM/HNK was investigated in BxPC-3 cells in vitro. The fluorescence intensity in the cells treated with SDC1-LB-MSN-Cou6 was 2-fold of LB-MSN-Cou6-treated cells, which was caused by SDC1/IGF1R-mediated endocytosis. As anticipated, its cytotoxicity was significantly increased. Furthermore, the mechanism was verified that SDC1-LB-MSN-HNK induced tumor cell apoptosis through the mitochondrial apoptosis pathway. Finally, the biodistribution, tumor growth inhibition, and preliminary safety studies were performed on BALB/c nude mice bearing BxPC-3 tumor models. The tumor growth inhibition index of SDC1-LB-MSN-GEM/HNK was 56.19%, which was 1.45-fold and 1.33-fold higher than that of the free GEM/HNK and LB-MSN-GEM/HNK treatment groups, respectively. As a result, SDC1-LB-MSN-GEM/HNK combined advantages of both GEM and HNK and simultaneously targeted and eliminated pancreatic cancerous and cancer-associated stromal cells. In summary, the present study demonstrated a new strategy of synergistic GEM and HNK to enhance the therapeutic effect of pancreatic cancer via the targeting depletion of tumor stroma.

RESUMO

The Tachinidae is one of the most speciose families in Diptera, and the exclusively parasitoid species play an important role in regulating populations of many herbivorous insects in ecosystems, including many agricultural pests. To better comprehend the characteristics and evolution of the mitochondrial genome for the Tachinidae, we are adding a massive amount of new molecular data by assembling the mitogenomes for 71 genera and 135 species from all four tachinid subfamilies through next-generation sequencing, and we are presenting the most comprehensive mitogenomic phylogenetic analysis of this family so far. Extensive rearrangements observed in the mitogenome of Admontia podomyia (Exoristinae) are unique for the entire suborder Cyclorrhapha. The rearrangement pattern suggests that the process involved a tandem duplication of the complete mitogenome, followed by both random and nonrandom loss of one copy of each gene. Additionally, five minor mitogenome rearrangements are discovered and described in three subfamilies. We present the largest species-level phylogenetic hypothesis for Tachinidae to date, based on mitogenomes of 152 species of Tachinidae, representing all four subfamilies and with five non-tachinid outgroups. Our analyses support the monophyly of the Tachinidae and most tribes and genera were recovered with good support, but the higher-level phylogenetic relationships within Tachinidae were poorly resolved, indicating that mitogenome data alone are not enough to unambiguously resolve the deeper phylogenetic relationships within Tachinidae.

RESUMO

China is conducting ecological restoration work in urban water bodies. Under anoxic and anaerobic conditions, pollutants transform and produce odorous and black substances, deteriorating the water quality, which is a significant problem in urban water bodies. Vallisneria natans has received widespread attention for its applications in water treatment and restoration. However, the efficiency by which V. natans reduces water pollution and allows sediment remediation requires further improvement. Therefore, in this study, we investigated the effect of V. natans coupled with carbon fiber on the restoration of water bodies and sediment compared with the control group that grew V. natans without carbon fiber. The oxidation-reduction potential (ORP) was selected as the main evaluation index for the water and sediment. Dissolved oxygen in the water and total organic carbon and total nitrogen (TN) in the sediment were also evaluated. V. natans coupled with carbon fiber significantly increased the ORP; that of surface sediment increased by 50 % and that of the water body increased by 60 % compared with the sediment without any bioremediation. Chemical oxygen demand, total phosphorous, and TN in water decreased by 61.2 %, 22.9 %, and 48.3 %, respectively. These results indicate that planting V. natans with carbon fiber can reduce pollutants in water (including humus) and sediments, effectively improving ORP in water and sediment.

RESUMO

Single materials that exhibit efficient and stable white-light emission are highly desirable for lighting applications. This paper reports a novel zero-dimensional perovskite, Rb4CdCl6:Sn2+, Mn2+, which demonstrates exceptional white-light properties including adjustable correlated color temperature, high color rendering index of up to 85, and near-unity photoluminescence quantum yield of 99%. Using a co-doping strategy involving Sn2+ and Mn2+, cyan-orange dual-band emission with complementary spectral ranges is activated by the self-trapped excitons and d-d transitions of the Sn2+ and Mn2+ centers in the Rb4CdCl6 host, respectively. Intriguingly, although Mn2+ ions doped in Rb4CdCl6 are difficult to excite, efficient Mn2+ emission can be realized through an ultra-high-efficient energy transfer between Sn2+ and Mn2+ via the formation of adjacent exchange-coupled Sn-Mn pairs. Benefiting from this efficient Dexter energy transfer process, the dual emission shares the same optimal excitation wavelengths of the Sn2+ centers and suppresses the non-radiative vibration relaxation significantly. Moreover, the relative intensities of the dual-emission components can be modulated flexibly by adjusting the fraction of the Sn2+ ions to the Sn-Mn pairs. This co-doping approach involving short-range energy transfer represents a promising avenue for achieving high-quality white light within a single material.

RESUMO

In the context of hydrogen production through water electrolysis, the development of efficient and stable electrocatalysts is of paramount importance. However, the creation of cost-effective electrocatalysts poses a significant challenge. In this study, a P and Nb co-doped NiFe2O4 nanosheet is designed and grown on Fe foam (referred to as P, Nb-NiFe2O4/FF). The P, Nb-NiFe2O4/FF exhibits a distinctive crystalline/amorphous heterostructure, and the co-doping of P and Nb in the material leads to the exposure of additional catalytic active sites, optimization of the electronic structure, and enhancement of charge conductivity. Additionally, the P, Nb-NiFe2O4/FF possesses a superhydrophilic surface for the enhancement of charge/mass transfer at interface and a superaerophobic surface, facilitating the efficient release of gas. The P, Nb-NiFe2O4/FF demonstrates remarkable oxygen evolution reaction (OER) and hydrogen evolution reaction (HER) activities, achieving overpotential as low as 247 mV and 127 mV, respectively, to attain the current density response of 100 mA cm-2. Based on the high bifunctional activities, the P, Nb-NiFe2O4/FF requires only a working voltage of 1.56 V to obtain the current density of 10 mA cm-2 in overall water splitting. Furthermore, the overall water splitting device of P, Nb-NiFe2O4/FF is integrated with a commercial solar cell to simulate a solar-powered water splitting system, resulting in as superior solar-to-hydrogen conversion efficiency of 15.11%.

RESUMO

Background: It is often challenging to select appropriate combination therapies to treat infections caused by carbapenem-resistant Enterobacterales (CRE) with high-level resistance to carbapenem. Methods: We investigated the in vitro synergistic activity of ceftazidime-avibactam-, polymyxin- or tigecycline-, and meropenem-based combinations using checkerboard assays against 16 CRE including Klebsiella pneumoniae carrying blaKPC-2 (CR1-blaKPC-2) and Enterobacter cloacae carrying blaNDM-1 (CR2-blaNDM-1) with meropenem MICs ≥128 mg/L. Time-kill assays were used to observe synergistic bactericidal activity. Results: Meropenem in combination with ertapenem, amikacin, tigecycline or polymyxin B, and tigecycline plus ceftazidime-avibactam showed weak synergistic activities against CR1-blaKPC-2 and CR2-blaNDM-1. Polymyxin B combined with tigecycline or ceftazidime-avibactam, and ceftazidime-avibactam plus amikacin showed synergistic effects against two tigecycline-non-susceptible KPC-producers or three ceftazidime-avibactam-resistant NDM-producer, and 50% (5/10) of strains with amikacin MICs ≥4096 mg/L, respectively. Synergistic interactions of ceftazidime-avibactam plus aztreonam or meropenem in checkerboard assays were measured for 100% (16/16) and 93.8% (15/16) of strains, respectively. The time-kill assay further verified that the ceftazidime-avibactam combination had the potential to restore aztreonam susceptibility and reduced meropenem MICs to 8 mg/L. Conclusion: Ceftazidime-avibactam plus aztreonam or meropenem could be an effective strategy for treating CRE infections, particularly those with high-level resistance to carbapenems and/or ceftazidime-avibactam.

RESUMO

Ecological floating bed coupled with microbial electrochemical system (ECOFB-MES) has great application potential in micro-polluted water remediation yet limited by low electron transfer efficiency on the microbial/electrode interface. Here, an innovative cathode-enhanced EOCFB-MES was constructed with nano-Fe3O4 modification and applied for in-situ remediation both at lab scale (6 L, 62-day operation) and demonstration scale (2300 m2, 1-year operation). The cathode-enhanced ECOFB-MES exhibited superior removal in TOC (81.43 ± 2.05%), TN (85.12% ± 1.46%) and TP (59.80 ± 2.27%), much better than those of original ECOFB-MES and anode-enhanced ECOFB-MES in the laboratory test. Meanwhile, cathode-enhanced ECOFB-MES boosted current output by 33% than that of original ECOFB-MES, which made a great contribution to the improvement of ectopic electronic compensation for pollutant decontamination. Notably, cathode-enhanced ECOFB-MES presented high efficiency, stability and durability in the demonstration test, and fulfilled the average concentration of COD (9.5 ± 2.81 mg/L), TN (1.00 ± 0.21 mg/L) and TP (0.10 ± 0.04 mg/L) of effluent water to meet the Grade III (GB 3838-2002) with stable operation stage. Based on the KOSIM calculation, the removal loads of cathode-enhanced ECOFB-MES in carbon, nitrogen and phosphorus could reach 37.14 g COD/(d·m2), 2.62 g TN/(d·m2) and 0.55 g TP/(d·m2), respectively. According to the analysis of microbial communities and functional genes, the cathode modified by Fe3O4 made a sensible enrichment in electroactive bacteria (EAB) and nitrogen-converting bacteria (NCB) as well as facilitated the functional genes expression in electron transfer and nitrogen metabolism, resulting in the synergistic removal of carbon in sediment and nitrite in water. This study provided a brandnew technique reference for in-situ remediation of surface water in practical application.

RESUMO

To improve the anti-metastasis effects of honokiol (HNK) on breast cancer, we designed cationic liposomes (Lip) in which HNK was encapsulated into Lip, and its surface was modified with negatively charged polysialic acid (PSA-Lip-HNK) for efficient treatment of breast cancer. PSA-Lip-HNK possessed a homogeneous spherical shape and high encapsulation efficiency. In vitro 4T1 cell experiments indicated that PSA-Lip-HNK increased cellular uptake and cytotoxicity via the endocytosis pathway mediated by PSA and selectin receptors. Furthermore, the significant antitumor metastasis impact of PSA-Lip-HNK was confirmed by wound healing and cell migration and invasion. Enhanced in vivo tumor accumulation of the PSA-Lip-HNK was observed in 4T1 tumor-bearing mice by living fluorescence imaging. For in vivo antitumor experiments using 4T1 tumor-bearing mice, PSA-Lip-HNK exhibited a higher tumor growth and metastasis inhibition compared with unmodified liposomes. Therefore, we believe that PSA-Lip-HNK well combined biocompatible PSA nano-delivery and chemotherapy, providing a promising drug delivery approach for metastatic breast cancer therapy.

RESUMO

KEY MESSAGE: The resistance of Huaidao5 results from the high constitutive expression of tolerance genes, while that of Huaidao9 is due to the cold-induced resistance in flag leaves and panicles. The regulation mechanism of rice seedlings' cold tolerance is relatively clear, and knowledge of its underlying mechanisms at the reproductive stage is limited. We performed differential expression and co-expression network analyses to transcriptomes from panicle and flag leaf tissues of a cold-tolerant cultivar (Huaidao5), and a sensitive cultivar (Huaidao9), under reproductive-stage cold stress. The results revealed that the expression levels of genes in stress-related pathways such as MAPK signaling pathway, diterpenoid biosynthesis, glutathione metabolism, plant-pathogen interaction and plant hormone signal transduction were constitutively highly expressed in Huaidao5, especially in panicles. Moreover, the Hudaidao5's panicle sample-specific (under cold) module contained some genes related to rice yield, such as GW5L, GGC2, SG1 and CTPS1. However, the resistance of Huaidao9 was derived from the induced resistance to cold in flag leaves and panicles. In the flag leaves, the responses included a series of stress response and signal transduction, while in the panicles nitrogen metabolism was severely affected, especially 66 endosperm-specific genes. Through integrating differential expression with co-expression networks, we predicted 161 candidate genes (79 cold-responsive genes common to both cultivars and 82 cold-tolerance genes associated with differences in cold tolerance between cultivars) potentially affecting cold response/tolerance, among which 85 (52.80%) were known to be cold-related genes. Moreover, 52 (65.82%) cold-responsive genes (e.g., TIFY11C, LSK1 and LPA) could be confirmed by previous transcriptome studies and 72 (87.80%) cold-tolerance genes (e.g., APX5, OsFbox17 and OsSTA109) were located within QTLs associated with cold tolerance. This study provides an efficient strategy for further discovery of mechanisms of cold tolerance in rice.

RESUMO

Sb3+-doped halide perovskites are promising candidates for solid-state lighting due to their diverse fluorescent colors and high efficiency. However, the mismatched high excitation energy with commercial UV chips is one of the critical issues to be addressed. Herein, a Bi3+ codoping strategy was established as a general and efficient approach to modulate the excitation spectrum from the Sb3+-doping center in all-inorganic perovskites of Cs2InCl5·H2O, Cs2NaInCl6, and Rb3InCl6. The incorporated Bi3+ greatly enhanced the splitting of the A band (1S0-3P1 transition) and boosts the enormous redshift of the low-energy branch in all these systems. The interactions persist strongly even at extremely low doping concentrations, suggesting a dipole-based long-range interaction. The results provide an in-depth insight into the contribution mechanism of Bi3+ to Sb3+ in all-inorganic perovskites, which throws light upon tuning the excitation spectrum of broadband emission from the extrinsic self-trapped exciton (STE).

RESUMO

With rapid worldwide spread of Coronavirus Disease 2019 (COVID-19), jointly identifying severe COVID-19 cases from mild ones and predicting the conversion time (from mild to severe) is essential to optimize the workflow and reduce the clinician's workload. In this study, we propose a novel framework for COVID-19 diagnosis, termed as Structural Attention Graph Neural Network (SAGNN), which can combine the multi-source information including features extracted from chest CT, latent lung structural distribution, and non-imaging patient information to conduct diagnosis of COVID-19 severity and predict the conversion time from mild to severe. Specifically, we first construct a graph to incorporate structural information of the lung and adopt graph attention network to iteratively update representations of lung segments. To distinguish different infection degrees of left and right lungs, we further introduce a structural attention mechanism. Finally, we introduce demographic information and develop a multi-task learning framework to jointly perform both tasks of classification and regression. Experiments are conducted on a real dataset with 1687 chest CT scans, which includes 1328 mild cases and 359 severe cases. Experimental results show that our method achieves the best classification (e.g., 86.86% in terms of Area Under Curve) and regression (e.g., 0.58 in terms of Correlation Coefficient) performance, compared with other comparison methods.

RESUMO

P-block metals have gradually been utilized to synthesize non-noble-metal catalysts for oxygen reduction reaction (ORR) due to the easily tunable localized p-orbitals and resulted versatile electronic structures. The high-density single-atom bismuth sites (Bi-NC) anchored onto nitrogen-doped three-dimensional porous carbon are proved to possess significant electrocatalytic ORR performance. Theoretical calculations unveil positively charged bismuth centers prominently improved the adsorption capacity of N-doped carbon to O2 . The p orbitals of Bi sites within Bi-NC easily generate hybrid states with p orbitals of O2 , thus promoting charge transfer and ultimately reducing the energy barrier of ORR. Benefiting from p-orbital electrons regulation of bismuth atoms, Bi-NC exhibit ORR half-wave potential of 0.86 V (vs RHE). Additionally, both liquid and quasi-solid zinc-air batteries with Bi-NC as air-cathodes achieve higher power density and specific capacity than 20 wt% Pt/C, and comparable stability and round-trip efficiency with 20 wt% Pt/C. The discovery sheds light on the theoretical and practical guidance for p-block metallic single-atom catalysts.

RESUMO

Organic-inorganic hybrid halides and their analogs that exhibit efficient broadband emission from self-trapped excitons (STEs) offers an unique pathway towards realization of highly efficient white light sources for lighting applications. An appropriate dilution of ns2 ions into a halide host is essential to produce auxiliary emissions. However, the realization of ns2 cation-based halides phosphor that can be excited by blue light-emitting diode (LED) is still rarely reported. In this study, a zero-dimensional Te-based single crystal (C8H20N)2TeCl6 was synthesized, which exhibits a yellow-orange emission centered at 600 nm with a full width at half maximum of 130 nm upon excitation under 437 nm. Intense electron-phonon coupling was confirmed in the (C8H20N)2TeCl6 single crystal and the light emitting mechanism is comprehensively discussed. The results of this study are pertinent to the emissive mechanism of Te-based hybrid halides and can facilitate discovery of unidentified metal halides with broadband excitation features.

RESUMO

Background: Whole-genome sequencing (WGS) has contributed significantly to advancements in machine learning methods for predicting antimicrobial resistance (AMR). However, the comparisons of different methods for AMR prediction without requiring prior knowledge of resistance remains to be conducted. Methods: We aimed to predict the minimum inhibitory concentrations (MICs) of 13 antimicrobial agents against Acinetobacter baumannii using three machine learning algorithms (random forest, support vector machine, and XGBoost) combined with k-mer features extracted from WGS data. Results: A cohort of 339 isolates was used for model construction. The average essential agreement and category agreement of the best models exceeded 90.90% (95%CI, 89.03-92.77%) and 95.29% (95%CI, 94.91-95.67%), respectively; the exceptions being levofloxacin, minocycline and imipenem. The very major error rates ranged from 0.0 to 5.71%. We applied feature selection pipelines to extract the top-ranked 11-mers to optimise training time and computing resources. This approach slightly improved the prediction performance and enabled us to obtain prediction results within 10 min. Notably, when employing these top-ranked 11-mers in an independent test dataset (120 isolates), we achieved an average accuracy of 0.96. Conclusion: Our study is the first to demonstrate that AMR prediction for A. baumannii using machine learning methods based on k-mer features has competitive performance over traditional workflows; hence, sequence-based AMR prediction and its application could be further promoted. The k-mer-based workflow developed in this study demonstrated high recall/sensitivity and specificity, making it a dependable tool for MIC prediction in clinical settings.

RESUMO

The increasing number of multidrug-resistant Gram-negative bacteria presents a serious threat to global health. However, colistin-resistant Acinetobacter junii has rarely been reported. We identified a colistin-resistant A. junii clinical isolate, AJ6079, in blood. The colony of AJ6079 presented a dry phenotype, and it was difficult to form a bacterial suspension, whilst transmission electron microscopy revealed that AJ6079 possessed a thick outer membrane. The phenotypic and genomic comparisons were conducted with one colistin-susceptible A. junii, which had the same antibiotic susceptibility profile except for colistin, and had the same KL25 capsule biosynthesis locus. The AJ6079 exhibited a slower growth rate, indicating that colistin-resistant A. junii possesses a higher fitness cost. The genome of AJ6079 had a G+C content of 38.7% and contained one 3,362,966 bp circular chromosome with no plasmid or mobile colistin resistance (mcr) gene. Comparative genomic analysis revealed that the AJ6079 contained several previously unreported point mutations in colistin-resistance-related genes involving amino acid substitutions in PmrB (N5K, G147C), LpxA (I107F, H131Y), and LpxD (F20I, K263R), which might be correlated with colistin resistance in A. junii. Further research is needed for verification as the genetic background was not exactly the same between the two isolates.

RESUMO

Macquartia Robineau-Desvoidy (Diptera: Tachinidae, Tachininae) represents one of the most ancient evolutionary lineages of tachinids, parasitizing Chrysomelidae larvae. We found four new Macquartia species collected by malaise traps, namely M. brunneisquama sp. nov., M. chinensis sp. nov., M. flavifemorata sp. nov., and M. flavipedicel sp. nov. These new species are described and illustrated, and their comparison with congeners as well as an identification key to the 12 species of Macquartia from China known to date are included. To determine the significance of the mitogenome architecture and evolution across different tachinid lineages of this primitive taxonomic group, four complete mitochondrial genomes were sequenced, annotated, and analyzed. The gene arrangements are consistent with the ancestral insect mitogenomes. The full-length sequences and protein-coding genes (PCGs) of the mitogenomes of the four species are all AT-biased. Analyses of Ka/Ks and overall p-genetic distance demonstrated that nad5 showed the highest evolutionary rate and nad1/nad4L were the most conserved genes among the four species. Phylogenetic reconstruction based on 13 PCGs strongly supported the monophyly of Macquartia, and the relationships of the four species are (M. flavifemorata + (M. flavipedicel + (M. brunneisquama + M. chinensis))). This study will help enhance our understanding of the taxonomic status and phylogenetic relationships in Tachinidae.

RESUMO

The sudden outbreak of coronavirus disease (COVID-19) triggered by SARS-CoV-2 infection has created a terrifying situation around the world. The spike protein of SARS-CoV-2 can act as an early biomarker for COVID-19. Therefore, controlling the spread of COVID-19 requires a low-cost, fast-response, and sensitive monitoring technique of spike protein. Herein, a photoelectrochemical (PEC) immunosensor for the detection of spike protein was constructed using the nanobody and an Mn (Ⅱ) modified graphitic carbon nitride (Mn/g-C3N4). The introduction of atomically dispersed Mn (Ⅱ) can accelerate the effective transfer and separation of photogenerated electron-hole pairs, which significantly boosts PEC performance of g-C3N4, thereby improving the detection sensitivity. As a recognition site, nanobody can achieve high-affinity binding to the spike protein, leading to a high sensitivity. The linear detection range of the proposed PEC immunosensor was 75 fg mL-1 to 150 pg mL-1, and the limit of detection was calculated to be 1.22 fg mL-1. This stable and feasible PEC immunosensor would be a promising diagnostic tool for sensitively detecting spike protein of SARS-CoV-2.

Sb3+ doping confers highly efficient and color-diverse broadband light emission to all-inorganic metal-halide perovskites. However, the emission mechanism is still under debate. Herein, a trace amount of Sb3+ ions (<0.1% atomic percentage) doping in the typical all-inorganic perovskites Cs2NaInCl6, Rb3InCl6, and Cs2InCl5·H2O allows universal observation of the fine structure in the photoluminescence excitation spectrum of the ns2 electron. A lifetime mapping method was utilized to reveal the origin of broadband emission triggered by Sb3+ doping, by which various fluorescence components can be differentiated. In particular, free-exciton emission was identified at the high-energy end of the broadband emission for all three doped systems. The excitation-energy- and temperature-dependent fluorescence decay further indicates the existence and origin of self-trapped states. The observed structural and vibrational symmetry-dependent emission behaviors suggest dipole interactions can dramatically alter Stokes-shift energy and modulate the light-emitting wavelength.
